Recommended Handling Instructions for Genomic DNA
1. Enclosed are samples of genomic DNA from the IHWG Cell and DNA Bank. Samples are labeled with the IHWG number as well as the cell line name. Each DNA sample was extracted from a standardized reference B-lymphoblastoid cell line from the Cell and DNA Bank inventory, the International Histocompatibility Working Group.
2. All DNA samples are in 1/10 strength TE buffer (1 mM Tris pH 7.5 and 0.1 mM EDTA).
3. The vials contain genomic DNA in the amount requested in the Order Form adjusted to a concentration of 100 ng/μL (nanograms per microliter). The exact amounts are listed in the included sample location map.
4. Upon receipt of the samples, it is suggested they be immediately logged into your laboratory inventory record system. Then:
a. Check each label to verify that all samples are present. Please contact us immediately with any question regarding sample identity or problems encountered with the shipment.
b. It is strongly suggested that the concentration of DNA in each sample be verified in your laboratory, according to your standard method, prior to testing. This will insure that no DNA has been lost during shipment.
c. “Quickspin” each tube to bring down the DNA droplets, then aliquot each sample to “working” tubes containing the desired amount of DNA for testing.
d. Freeze all tubes that will not be used immediately at -20° C or lower. DNA to be used within 2 weeks may be stored at 4° C. Avoid frequent freeze-thawing of individual DNA samples.
